  • Postpartum depression, saffron vs fluoxetine · 30 mg over 6 weeks (limited evidence) · Primary endpoint missed — 15 mg twice daily against fluoxetine 20 mg twice daily in mild-to-moderate postpartum depression. There was no significant difference between arms, and complete response was 40.6% on saffron vs 50.0% on fluoxetine — an active-comparator trial with no placebo arm, so it does not establish that saffron beat nothing.

  • Depression meta-analyses (pooled) · 30 mg (limited evidence) · Primary endpoint missed — Pooled saffron-vs-placebo effect was large but the confidence interval reached zero (SMD −0.86, 95% CI −1.73 to 0.00), and saffron did not differ from fluoxetine. Eight small trials, most from a single country. Treat 30 mg/day as the dose those trials used, not as a settled effective dose.

Label reading notes

  • Almost every trial used 30 mg per day of a standardised extract — check whether the label means extract or dried stigma powder.
  • Branded standardised extracts (affron, Safr'Inside) are dosed at 28-30 mg; raw powder blends at the same milligram count are not the same material.
  • Trials ran 4-6 weeks in people already on an antidepressant, under clinician care. Do not stop or change a prescription to try this.
