Trial floors
Primary
Stress and cortisol RCT (Chandrasekhar 2012) · 300 mg over 8 weeks · Benefit shown — High-concentration full-spectrum root extract, 300 mg per day for eight weeks in a randomized double-blind placebo-controlled trial of 64 adults. Serum cortisol fell significantly versus placebo and a stress scale improved. The reference point for label comparison.
Secondary
Dose-ranging trial (Salve 2019) · 250–600 mg over 8 weeks · Benefit shown — Three-arm trial comparing 250 and 600 mg per day of root extract. Perceived stress improved on both doses and cortisol on both, with the stronger signal at 600 mg. Establishes the studied range rather than one number.
Safety red flags
- Not for pregnancy or breastfeeding without a clinician's explicit ok; ask first with autoimmune disease or an overactive thyroid.
- On sedatives, thyroid medication, or immunosuppressants: tell your prescriber before starting.
Label reading notes
- Compare the label with 300-600 mg per day of standardized root extract; "ashwagandha" with no extract or standardization statement is not a comparable quantity.
- The shelf runs from 100 mg inside blends to 1200+ mg standalone; most of it sits outside the honest comparison range.
- Safety profile in randomized trials is clean (Verma 2021, PMID 33338583, tracked in our Nutrafol guide).
- Common caution block: pregnancy, autoimmune conditions, hyperthyroidism — clinician first.
